Virus-induced Gene Silencing

Virus-induced gene silencing (VIGS) was first used to describe the recovery phenomenon of virus-infected plants, it is an RNA-mediated antiviral defense mechanism of plant, and has been developed as a reverse genetics tool for gene function analysis recently. VIGS refers to the virus carrying the target gene fragment infecting the plant, which can induce the silencing of homologous genes in the plant, thereby helping researchers to analyze the function of the target gene through the study of plant phenotype changes. VIGS includes three important steps, inserting the target gene fragments to be silenced into the viral genome, infecting a suitable host plant, and finally using the plant's defense mechanism against virus infection to silence the target gene. Compared with traditional methods of studying plant gene blocking such as gene knockout, and antisense suppression, VIGS has the characteristics of simplicity, efficiency, short periodicity, no need to transform plants, and the ability to silence gene families, etc., and its silencing effect can be passed on to future generations under certain conditions. Therefore, VIGS has been widely used in the functional identification of genes related to physiological pathways such as plant disease resistance and stress resistance, growth and development, and metabolic regulation.

  • The most common use of VIGS technology is to identify plant functional genes. VIGS can induce target gene silencing in plant tissues and organs. VIGS technology is a fast and effective method for studying plant gene function, especially for some non-model plant gene functions that are difficult to transform.
