Phage Display Library Screening
Phage display is a biological technology that inserts the DNA sequence of a foreign protein or polypeptide into the appropriate position of the structure gene of the phage coat protein, so that the foreign gene or polypeptide can be expressed along with the expression of the coat protein. At the same time, the foreign protein can be displayed on the phage surface following the reassembly of the phage.
After the phage library is constructed, it is necessary to screen recombinant phages that meet the requirements. Phage library screening can be performed in vivo or in vitro. In vivo phage display peptide library screening is performed in living animals, because of the heterogeneity of vascular molecular endothelium, phages can be selectively directed to different tissues, so that phage display peptides specific to tissues can be screened. In vitro screening is a biological washing procedure. Because of the large library capacity of phage libraries, it is necessary to have specific and efficient screening methods to screen the fragments which have high specificity but low occurrence rate and low affinity in these libraries. At present, biological washing method is commonly used, so we focus on this method.
Bacteria, usually Escherichia coli is used to load phage libraries and helper phages, then target protein or peptide is expressed on the phage protein coat. Recombinant phages are incubated with a stationary phase in which specific screening molecules have been added, so that proteins or peptides that bind specifically do not wash off, while proteins that do not bind specifically are washed off. The conjugated recombinant phages are then eluted and re-infected with the bacteria, with serval cycles, enough recombinant phages are produced so that they can meet the requirement of customers.
Screen Steps
- Assembly. Phagemid library and helper phage are assembly into E. coli.
- Incubation. Assembled phages are incubation with specific surface.
- Binding. Bound phages do not wash off, while unbound phages are missing.
- Elution. Bound phages can elute to re-infection.
