Phage Display

The principle of phage display technology is to insert the DNA sequence of foreign protein or peptide into the appropriate position of the structural gene of phage coat protein, so that the exogenous gene can be expressed with the expression of coat protein without affecting the normal function of coat protein. Finally, the foreign protein or peptide is displayed on the phage surface in the form of fusion with coat protein. The technology is primarily used to display interested peptides, antibodies, scaffolds or other proteins on the surface of phage and then to screen constructed libraries from a variety of displayed phages. This technology establishes a direct physical link between the protein and the gene, allowing rapid screening of required antibodies.

First step for phage display is library construction. Sequences of nucleic acid used to construct the library can be synthesized artificially or by cDNA. cDNA is usually used to obtain sequence for library construction. Phage display library construction process is to extract mRNA, obtain template cDNA by reverse transcription, and then design appropriate primer PCR to obtain the required gene sequence. This gene is then cloned into a phage vector and transformed into a receptive cell with phage helper to form a display library. The main phage display systems used to build libraries are the single-stranded filamentous phage display system, λ phage display system, T4 phage display system and T7 phage display system. According to different target, libraries can be classified with cDNA library, antibody library, protein library and random peptide library.
