Recombination Virus-like Particles

Virus is a non-cellular form composed of nucleic acid molecule (DNA or RNA) and protein, it also called viral particles. Viral particles can use the host's cell system to replicate themselves, but cannot grow and replicate independently. Recombination viruses are products obtained by genetic modification of viruses, for example, knocking out or inserting a gene to get viruses with reduced toxicity.  Recombination virus-like particles (rVLPs) are nanostructures that resemble the structures of viruses. A key factor regarding rVLPs safety is the lack of a viral genome, which enhances safety during both manufacture and administration. The rVLPs can be used as a preventive viral vaccine with improved safety to cause a high degree of humoral immunity and cellular immunity. Besides, rVLPs can also be targeted to specific sites as a delivery system. Due to the advantages compared to other types of vaccines, the interest in rVLPs technology has increased in recent years. There are many production platforms to amplify the rVLPs. The selection of expression systems for rVLPs production must take into account issues such as protein folding and post-translational modification. These expression systems include bacteria, yeast, insect cells, plant, etc.

Amplifying Recombinant Viruses in Bacteriaand Yeast

Bacteria and yeast are both easily scaled up and cost- effective production platforms. Especially for bacteria, bacteria have a short reproduction cycle, low cultivation cost and easy operation, so bacteria are the most suitable system for overexpression of many proteins of interest. They are also suitable for rVLPs amplification. Yeast represents a step forward in rVLPs production because of the ability to perform post-translational modifications. So yeast is more suitable for eukaryotic recombination viruses.

Amplifying Recombinant Viruses in Baculovirus/insect Cell (B/IC) Expression System

B/IC expression system includes two phases, which are infection phase and production phase. Insect cells are easily infected by baculovirus, and they are a suitable production platform for large-scale amplification of baculovirus. Moreover, Insect cells are easier to culture than other cells. When the insect cells are in high density culture condition, they can produce a large amount of rVLPs that are properly folded.

Amplifying Recombinant Viruses in Plant Cells

Plant-based expression systems have been welcome systems for rVLPs production in recent years. Plant-based production platform has these advantages, such as, easy to express target rVLPs, produce complex proteins, low risks of contamination with human or animal pathogens, etc. Therefore, plant-based production platform represents an attractive alternative to the traditional expression systems. Plant cell suspension as well as plant tissue cultures have been exploited for the production of valuable secondary metabolites, in recent years, they have also been developed for the production of rVLPs. Several examples are available of rVLPs production in plants, such as HPV type 16 or influenza.
