Retroviruses

Retroviruses are a class of single-stranded RNA viruses that contain reverse transcriptase. Viral RNA is used as the template to synthesize cDNA under the function of reverse transcriptase, and then particles are formed through DNA replication, transcription, translation and other processes. The structure of retroviruses includes three parts, core proteins coded by gag gene, reverse transcriptase coded by pol gene and capsule glycoprotein coded by env gene. Retroviruses expression system is an efficient and novel recombinant protein expression system, which consists of a reverse transcription expression vector, an auxiliary vector (a protein needed for expressing viral packaging) and a packaging cell line. However, retroviruses can only infect dividing cells, such as embryonic stem cell (ESC), neural stem cell (NSC) and hematopoietic stem cell (HSC).

  • MMLV Retrovirus Packaging
  • The vector of MMLV (moloney murine leukemia virus) retrovirus, is an efficient tool which can induce exogenous gene into dividing cell permanently. Even belonging to the same family, MMLV retrovirus is different from lentivirus which can infect both dividing and non-dividing cells, it can only infect dividing cells. As a method of gene transmission, MMLV retrovirus vectors are particularly effective in producing iPS cells. Creative BioMart Vir-Sci provide two kinds of packaging services, MMLV retrovirus and self-inactivation MMLV retrovirus. We have developed a range of proprietary technologies and reagents that have significantly improved the MMLV retrovirus packaging process in terms of virus titer, purity, activity and consistency, especially for the MMLV retrovirus vectors used in our vector construction services. Combining the design platform with the vector and virus production line, we are able to provide high-quality customized MMLV retrovirus products.

  • MSCV Retrovirus Packaging
  • The vector of MSCV (murine stem cell virus) retrovirus is often used as an effective transgenic tool to express exogenous genes in embryonic stem cells (ES cells), hematopoietic stem cells (HS cells) and other mammalian cell lines. The 5 'LTR of MSCV retrovirus vectors use PCMV virus sequence of mouse stem cells, after it has been designed carefully, it can activate the expression of exogenous genes in pluripotent stem cells, thus overcoming the problem of gene silencing. After optimization, the vectors have a high copy number in E. coli, packaged retroviruses have high titer and highly efficient transduction ability to most host cells, and can effectively integrate vectors into the target cell genome and achieve high levels of expression of exogenous genes. Due to technical advantages, our MSCV retroviruses packaging is higher in quality, more efficient and more affordable.

Technology Advantage

  • Stable integration of exogenous genes. The target genes transduced by retroviruses can be stably integrated into the genome of host cell and thus be stably inherited followed by the host cell division.
  • Broad host range. Our retroviruses packaging contains VSV-G envelope proteins that have a wide range of affinity for transduction from mammalian cells such as human, mouse and rat cells and other types of cells.
  • Large vector capacity. The retrovirus genome is about 8 KB in size. In our retroviruses vector, the essential components for virus packaging and transduction are about 2.5 KB, with the remaining 5.5 KB for the target sequence.
  • High level gene expression. The 5'LTR of retrovirus vector contains a very strong promoter that enables high expression of the target gene.
  • Uniform gene copies. Normally, our retrovirus transduction is able to transfer foreign genes into target cells uniformly.
