Viral Vector Delivery

Gene delivery is the introduction of genes into cells, and these cells then produce the required therapeutic substances, mainly protein products. For viral delivery, foreign genes are attached to the virions that have been modified to remove harmful parts and introduced into cells to achieve gene transmission. All viruses can be developed into a tool for gene transmission through a series of processes, such as deleting genes related to carcinogenesis, toxicity and replication, and inserting foreign therapeutic genes at appropriate locations. However, due to the wide differences in the life cycle and molecular pathology of different viruses, four types of viral vectors are mainly used: retroviral vectors (including lentiviral vectors), adenovirus vectors, adeno-associated virus vectors, and simple herpesvirus vector.

Gene Delivery Method

The delivery of genes to host cells involves the following two steps:

  1. Delete some of the essential genes of the virus, and replace the non-essential genes of the virus with the foreign genes (target genes) (the non-coding region remains unchanged)
  2. Recombinant vector plasmid

The foreign gene, cis-acting element and plasmid backbone are packaged into the viral capsid under the action of an auxiliary system (trans-acting element) to form a recombinant vector plasmid.

Applications

In recent years, substantial advances have been made in the development of technologies that allow specific cells to be targeted for several types of vectors, especially viral vectors. These vectors have been used in 70% of gene therapy clinical trials. Specific viruses were selected as gene delivery vehicles because of their ability to carry foreign genes and the ability to effectively deliver these genes in relation to effective gene expression.
