We first designed and cloned primers of appropriate length based on the full-length sequence of the CP gene of the virus, and evaluated the designed primers. Then we extract the RNA of the plant virus, and then obtain cDNA by reverse transcription. We use cDNA as a template to amplify the designed primers by PCR. Then recover and sequence the amplified product and compare it with the full sequence of the CP gene.
We use overlap-PCR technology to splice the CP gene fragment and the target gene to construct a plant expression vector. Then we identified the constructed plant viral vectors.
