Baculovirus Expression System

The baculovirus expression system is a popular method for the production of recombinant proteins. Baculovirus shuttle vector (bacmid) contains a mini-F replicon, which allows autonomous replication and stable segregation of plasmids at low copy number, a selectable kanamycin resistance marker, and attTn7, the target site for the bacterial transposon Tn7, all inserted into the polyhedrin locus of Autographa califomica nuclear polyhedrosis virus (AcNPV). Bacmid can replicate in E. coli as a large plasmid, and it is still infectious after being introduced into insect cells. Recombinant bacmid has been widely used for protein expression of viruses, fungi, plants, and animals.

The donor baculovirus vector with target gene is transformed into E. coli DH10Bac. Bacmid with transposition was inactivated due to the insertion of LacZ gene, making recombinant bacmid colony colorless, then recombinant bacmid DNA was extracted and used for transfection of insect cells Sf9.

Technology Advantage

  • Easy to operate. Baculovirus genome is small, with relatively simple molecular biological characteristics and a variety of restriction enzyme sites.
  • Baculovirus virions are rod-shaped and have the plasticity to accommodate large foreign genes.
  • p10 and polyhedron in baculovirus are highly transcribed during the late stages, their promoter can express the target protein efficiently.
  • The host of baculovirus is insect cells, and the target protein can be processed and modified by the post-translational modification system of insect cells.
  • High security. Baculovirus expression vector is very specific and can only replicate in insect cells.
  • Compared with mammalian cells, the cell lines of insect culture are relatively easy to grow, and the suspension culture of insect cells can produce a large number of target proteins.
