Baculovirus Vector Systems

Baculovirus vector systems are widely used to express recombinant proteins in cultured insect cells. Many eukaryotic proteins undergo posttranslational modifications (such as glycosylation) that occur only in eukaryotic cells, or they require eukaryotic cell environments (such as membrane proteins) for proper folding. In these cases, prokaryotic expression systems are often inadequate, and baculovirus expression systems may be a good choice. Currently the most widely used baculovirus expression system is a split virus of Autographa california multiply enveloped nuclear polyhedrosisvirus (AcMNPV), which is referred to as baculovirus for short. These vectors are characterized by: baculovirus expression systems all use protein modification, processing, and transport systems that exist in higher eukaryotic cells, which are eukaryotic expression systems; AcMNPV is a non-helper virus and does not require any auxiliary factors. Insect cells suitable for suspension growth proliferate in large numbers, so it is convenient to express a large number of recombinant proteins; the expression system makes the expression product in a dissolved state; the baculovirus gene is large (130kb), which is suitable for cloning large fragments of foreign genes.

Our Manufacturing Process of Baculovirus Vector

  • The gene of interest was cloned into the pBV vector under the control of a strong promoter.
  • The second step is to transform into a baculovirus, which is actually a very large plasmid containing the baculovirus genome, and can be modified to carry the lacZ gene and the attTn7 docking site inserted into the lacZ coding region.
  • The purified baculovirus plasmid DNA can be used to transfect insect cells to produce baculovirus, which can be used to produce the recombinant protein of interest. The most common cell line for expressing recombinant proteins from rod-shaped vectors is Sf9. The cell line is suitable for a variety of culture and medium conditions, including suspension or monolayer cultures and serum-free medium.

Advantages

  • Eukaryotic system. Post-translational processing of proteins by insect cells is similar to mammalian cells. Therefore, our system is particularly suitable for expressing mammalian and other eukaryotic proteins whose functions require proper post-translational processing, such as covalent modification or membrane targeting, which is not present in the prokaryotic expression system.
  • Strong expression and good solubility. In most cases, the target protein is highly expressed, soluble, and can be easily recovered from infected cells.
  • Easy to scale up. In our system, baculovirus obtained from the initial transfection of insect cells can be used to infect more cells to further expand the virus titer. Therefore, our system of protein production can be scaled up reproducibly.
  • High efficiency. The system can express proteins with post-translational processing characteristics of eukaryotic cells and good adaptability for large-scale applications.
